The only way to unlock Aunt May is to do Deadpool's last mission, meaning you have to first play all the Deadpool missions, get the red bricks, buy all the extras, then you unlock Deadpool. Then you have to complete the first two Deadpool side quest then do the third. Aunt May is the hardest character to unlock in Lego Marvel!
