Have a power braclet item card and use it on the lone tree in that sqare! Then clear the water temple scenario as Darunia and beat Ruto to get her!

That requires a search. Get a compass, and check where the secret is. When you find out, get the item you need to explore the secret area. When you find the new reward, then Ruto is capable of being unlocked. Beat the mission, (Rank doesn't matter) and you will unlock Ruto.
