Any time from Chapter 4 onward. He's always in Vampire castle, but the fight gets considerably harder in Chapter 7.

Hmm, but it always says something about six objects I would have to put into the door in order to open it, but up until now I haven't ever had any further clue about them. I'm always walking through the villages, but no one seems to know about these "keys". :/

I don't think the game gives much in terms of clues. They're the six obelisk like things you find around the world map, like outside of Lontano Villa. Be prepared for a boss fight at each.
